Sesamin, one of the lignans presents most abundantly in sesame,is scientifically proven to protect and enhance liver functions, maintain healthy blood pressure, lower harmful blood cholesterol levels and serve as an antioxidant.

Sesamin is supplied as a crystalline solid. A stock solution may be made by dissolving the sesamin in an organic
solvent purged with an inert gas. Sesamin is soluble in organic solvents such as ethanol, DMSO, and dimethyl formamide.The solubility of sesamin in these solvents is 0.5, 12, and 33 mg/ml, respectively. Sesamin will be stable for at least six months in these solvents if stored at -20°C.

Sesamin is sparingly soluble in aqueous buffers. Further dilutions of the stock solution into aqueous buffers or isotonic saline should be made prior to performing biological experiments. For maximum solubility in aqueous buffers, sesamin should first be dissolved in DMF and then diluted with the aqueous buffer of choice. Sesamin has a solubility of 50 µg/ml in a 1:10 solution of DMF:PBS (pH 7.2) using this method. Store aqueous solutions of sesamin on ice and use within 12 hours of preparation. Although the aqueous solutions of sesamin may be stable for more than 12 hours, we strongly recommend using a fresh preparation each day.

Sesamin is a non-competitive inhibitor of ?5-desaturase. It inhibits the conversion of dihomo-?-linolenic acid to arachidonic acid with a Ki value of 155 µM in rat liver microsomes.1

Metabolic Role:

Japanese Scientists' studies clearly demonstrated that sesamin markedly influences the hepatic fatty acid oxidation and synthesis in the rat. Sesamin appears to be an active PPAR ligand and the most potent inducer of hepatic fatty acid oxidation among the various naturally occurring compounds so far reported. Moreover, the stutides showed that sesamin decreases the hepatic lipogenic enzyme gene expression through the down-regulation of SREBP-1. The changes in the hepatic fatty acid metabolism by dietary sesamin were associated with a decrease in triacylglycerol secretion by perfused rat liver. Therefore, the reduction in the hepatic lipoprotein production may account for the serum lipid-lowering effect of dietary sesamin. The studies also developed sesame seed lines rich in sesamin and sesamolin that enhance hepatic fatty acid oxidation and lower the serum triacylglycerol level more than a conventional cultivar. The studies using rats provided the scientific basis to demonstrate the health-promoting characteristics of sesame seeds and oil. The consumption of sesamin is expected to enable to improve the lipid metabolism and hence be beneficial to health by preventing arteriosclerosis in humans also.
